Your original bet, in your words, was if trump is the Republican nominee, he will win in a landslide. There was neither mention in either of those original bets that the Democratic nominee had to be Biden. The bet was only about trump being the candidate and winning in a landslide.
Only later, after you offered the original bet multiple times and had it accepted, did you then try changing it to including Biden. But the original bet had been offered and accepted. Further, you later referenced the "original" bet being the one that was live.
So unless there is other evidence, the ruling is final, and based on the evidence in the link, there is really no way to argue out of that.
You haven't lost until trump doesn't win in a landslide. E.T. hasn't lost based on Biden's resignation, since the original bet never assigned Biden to E.T.
